Ferulate-sinapyl alcohol cross-coupling reaction improves the understanding of grass cell wall lignification

چکیده
During the lignification of grass cell walls, ferulate acts like a nucleation site that can cross couple with monolignols, i.e. coniferyl alcohol and sinapyl alcohol, to form dimeric products. Here we investigated in detail ferulate-sinapyl cross-coupling products an effort on better understanding nature scope lignin-ferulate interactions. In biomimetic oxidative system, (SA) reacted ethyl (FA), simplified model for feruloyl polysaccharides plants, give mixture dozens We delineated structure each product by GC-MS NMR characterizations. It was found readily reacts producing various cross-coupled dimers including SA-(β–5)-FA (compound 6), SA-(β–8)-FA (both ester lactones, compounds 7 9), SA-(β–O–4)-FA 8) addition homo-coupled from or alcohol. 8 identified first time vitro free radical system SA-(β–O–4)-SA 3 accurately quantified as less than 5%, while half-open syringaresinol 5 discovered when dimerization SA catalyzed peroxidase buffer system. Moreover, one new compound 18 formed linkage (de-protonated) NMR, possible FA-(8–β)-SA dimer 19* proposed.

Preparation and relevance of a cross-coupling product between sinapyl alcohol and sinapyl p-hydroxybenzoate.
Cross-coupling of sinapyl p-hydroxybenzoate and sinapyl alcohol produces an 8-8-cross-coupled product that is also detected in lignifying poplar tissues, implicating sinapyl p-hydroxybenzoate as a lignin precursor.

The cell walls of grasses such as wheat, maize, rice, and sugar cane, contain large amounts of ferulate that is ester-linked to the cell wall polysaccharide glucuronoarabinoxylan (GAX). This ferulate is considered to limit the digestibility of polysaccharide in grass biomass as it forms covalent linkages between polysaccharide and lignin components. Candidate genes within a grass-specific clade...

Lignification limits grass cell-wall digestion by herbivores. Lignification is spatially and temporally regulated, and lignin characteristics differ between cell walls, plant tissues, and plant parts. Grass lignins are anchored within walls by ferulate and diferulate cross-links, p-coumarate cyclodimers, and possibly benzyl ester and ether cross-links. Cell-wall degradability is regulated by li...
متن کاملPlant cell wall lignification and monolignol metabolism
Plants are built of various specialized cell types that differ in their cell wall composition and structure. The cell walls of certain tissues (xylem, sclerenchyma) are characterized by the presence of the heterogeneous lignin polymer that plays an essential role in their physiology. This phenolic polymer is composed of different monomeric units - the monolignols - that are linked together by s...
